You draw a distinction between retest date and expiry date and then say suppliers use the wrong word. Which word do you think they should use, given that most of them have no study behind either?

Retest, with a stated interval and a note that no formal stability study supports it. That is an honest description of a chemical supplier’s position and it is standard practice in the wider chemical trade. Printing expiry implies a study exists, which is the specific inference we object to.

Your piece assumes readers have a clinician ordering panels for them. A great many of us are buying our own, from services that supply an interval, a flag and nothing else. The apparatus you describe is not available to us at all.

Your article argues for two orthogonal methods and reports the lower figure, but the people running a single twelve-minute method have a cost story you do not address. A full orthogonal pair doubles the turnaround and at least doubles the cost, which is why the market does not do it. The criticism of method disclosure is fair. The criticism that a single method is wrong is unfair to the constraints people operate under.

We are careful to say that a second method costs instrument time on a sample already in the autosampler, which is substantially less than twice the turnaround, but you are right that we underweight the commercial reality that a buyer setting a budget for testing is trading thoroughness for speed and price. Where we would push back is that those constraints are not technical or regulatory ones. They are market ones, and markets can change if enough buyers demand it.

Sub-two-micron columns changed everything about what is practical for peptide separations, but I would push back on the statement that particle size gains are costless. The pressure limit of most commercial instruments is three hundred bar, and trying to force 1.7-micron particles at eight millilitres per minute on a 4.6-millimetre column will send you there quickly. Peak capacity is not free.

Correct on the pressure cost, and that belongs in the method section. The trade-off is real, which is why many laboratories use core-shell superficially porous particles as a compromise: they are substantially cheaper than sub-two-micron packings, deliver most of the efficiency gain at a lower pressure, and the efficiency-cost-pressure triangle is the actual business decision people make. We should have named it.

One practical note from the other side of the counter. When a customer asks us for the gradient we send it. When a customer asks a reseller, the reseller does not have it, because they were sent a PDF with a number on it. The gap you are describing is often two links down the chain rather than at the laboratory.

If the laboratory report travels intact instead of being transcribed into a house certificate, the method travels with it. Your table of what each method can see puts "only if resolved" against isoaspartate for RP-HPLC. That understates the difficulty. Resolving isoAsp from Asp routinely requires a method developed for the purpose, and on a generic gradient the two are frequently indistinguishable even at forty minutes.

A tyre pressure gauge is silent on tread depth, brake pads and the driver, and it is still the right instrument for its question. The failure is in a market that owns one gauge and calls the reading roadworthiness.

What is missing is not evidence that it occurs but evidence about how much of it is present in any particular commercial vial, which is a different absence and the one we should have named.

There is no randomised evidence that treating the low-T3 pattern of energy restriction improves any outcome, and there is a mechanistic argument that suppressing an adaptive response is unlikely to help. We report it as adaptation for those reasons and would report a trial that changed the picture.

My HbA1c was 6.1 before starting and 6.0 after four months. My clinic recorded this as no improvement in glycaemic control. My continuous monitor says my average glucose fell by a fifth over the same period. Which is measuring what?

Both are measuring correctly and the discrepancy is worth pursuing with your clinic rather than with us. A 0.1-point change is well inside the reference change value for HbA1c, so the assay has not detected a change; whether that is because the change is genuinely small or because something is affecting your glycation is not answerable from the numbers alone.

A handheld device measures impedance across the upper body and infers the rest, and the inference is least reliable exactly where you sit: older, substantial weight change, changing hydration. Twenty-four months is a common default and “industry standard” is an accurate description of the practice rather than a justification of the claim. The distinction we would press is between an expiry date, which asserts shelf life, and a retest date, which asserts only a review interval. The second is defensible without stability data. The first is not.

A longer needle in a heavier person is not more likely to reach the right layer; it is only more likely to go past it in a thinner limb. We would put the ultrasound measurement studies in front of your nurse rather than argue from authority.

Gradient, by roughly two to one in our four conditions: holding the threshold at 0.10 per cent, lengthening the gradient cost 0.9 points, while holding the gradient and tightening the threshold cost 0.4 to 0.9 depending on which gradient. We should have printed that decomposition in the table and it now appears in the note. If a supplier will disclose only one value, it should be the gradient.

Your submission design has a hole in it. Eight vials from one lot cannot separate variation between laboratories from variation between vials, because you have no replicate within a laboratory to estimate the second. Two vials each is a start and it is not enough, and the honest conclusion from your table is that the four figures differ, not that the laboratories do.

Correct, and the criticism is well aimed. With pairs we can see within-laboratory agreement, which was good in every case, but we cannot decompose the remaining variance properly. The four-condition study on a single sample was designed to isolate the method effect for exactly that reason, and it is the stronger half of the exercise. We should have said which half carried the weight.

We stopped short because the magnitude is formulation-specific and because most reconstituted research vials are in unbuffered water or bacteriostatic water, where the argument is about the interface rather than about pH. The claim of end-to-end control is the thing we criticise, and we do criticise it, in the article and again in the closing. You write that leucine and isoleucine cannot be distinguished by tandem mass spectrometry. That is too absolute. Side-chain fragmentation under high-energy conditions produces diagnostic w and d ions, and the discrimination has been demonstrated repeatedly.

I run analytical services and I would put it differently: we report what we measured, in the words our clients ask for. If the Journal wants the word confirmed retired, write to the buyers, not to us.

I would add one omission to your six lines: the date and nature of the last calibration. A parts-per-million figure from an instrument last calibrated a fortnight ago is a different claim from one calibrated that morning with an internal standard.

The claim that a reproduced spectrum is worth more than any number in the document seems overstated. Most buyers cannot read a spectrum, and a printed image invites false confidence rather than scrutiny.

Worth saying more plainly: a purity certificate and a sterility assurance are not merely different tests, they are different disciplines with different facilities, and no amount of chromatography will ever bear on it.
