A modest proposal: publish the shipping cost alongside the result. It sounds irrelevant and it is the fastest way for a reader to tell a genuine retail purchase from a sample that arrived by arrangement.

Community-funded testing has solved the money problem in a handful of cases, and it introduces a new one: the group choosing the target has its own preferences about the answer. It is a better arrangement than seller-funded testing and it is not neutral.

You treat aggregation as a degradation pathway alongside the chemical ones. It is worth separating, because it is physical, it is often reversible in principle and irreversible in practice, and it is the one pathway that reversed-phase chromatography will not report on at all.

Single-use indicators are much better than nothing and much weaker than a logger, because they tell you a threshold was crossed and not for how long. Ten minutes above eight degrees on a loading bay and eleven hours in a hot van produce the same red mark.

A logger recording every five minutes and a logger recording hourly will produce different excursion findings on the same journey, because the short spike at the depot falls between readings. Sampling interval is a specification and it is almost never quoted with the logger data.

It is available, with effort: published biological variation databases are open, the reference change value is one line of arithmetic, and your own previous result is the comparator that does most of the work. But you are right that the market supplying these panels has no incentive to include any of it, and that is worth stating.

The economic point is missing from your analysis. A second orthogonal determination roughly doubles the cost of knowing, and the buyers in this market are mostly individuals paying out of pocket. Recommending it without saying what it costs is the kind of advice that gets ignored rather than argued with.

Capillary electrophoresis is the forgotten answer here. It separates on charge-to-size in free solution, it needs a few nanolitres, and for anybody who already has the instrument it is an afternoon. The barrier is not cost or difficulty; it is that hardly anyone in this trade has been trained on it.

On the section about diode-array detection and peak purity, I would add that true peak purity assessment requires library matching or at least spectral comparison across the peak width. A homogeneous spectrum tells you the peak is probably pure. A spectrum that shifts across the peak tells you it is not, and that information closes a gap the article identifies correctly.

Your worked example varies gradient and threshold together and reports a 1.8-point spread. Which of the two contributed more? The article does not say, and the answer matters for what you are asking suppliers to disclose first.

Gradient, by roughly two to one in our four conditions: holding the threshold at 0.10 per cent, lengthening the gradient cost 0.9 points, while holding the gradient and tightening the threshold cost 0.4 to 0.9 depending on which gradient. We should have printed that decomposition in the table and it now appears in the note. If a supplier will disclose only one value, it should be the gradient.

One request for the checklist. Ask whether the stability data supporting the storage statement was generated on this formulation or on the same peptide in a different one. The distinction decides whether the number applies to the vial in your hand.

A reference interval is a description of a population, not a definition of health, and your piece is one of the few places I have seen that said so plainly. Biological variation is the concept I would add. Each analyte has a characteristic within-person variability, and for some of them a change has to be substantial before it means anything at all. Without that figure a delta is uninterpretable, and it is available in published tables.

Anything that shortens red cell survival lowers the result independently of glycaemia. Haemoglobin variants interfere with some assay methods and not others. These are not exotic situations, and a value that does not fit the clinical picture deserves a different measurement rather than a firmer conclusion.

Fasting glucose and glycated haemoglobin can move in opposite directions over a short period and both be correct, because they describe different windows. Your table makes that visible in a way I have not seen elsewhere and I have borrowed it for teaching.

That is the correct reading, and the unmeasurable part is the honest part. We would add only that vendor-published reports are not worthless — a vendor willing to commission testing at all is behaving better than one that will not — they are simply weaker in a specific way.

The half-life to steady-state arithmetic deserves to be spelled out every time, because it is the single most useful calculation in this subject. Four to five half-lives to approach steady state means a weekly molecule with a week-long half-life takes over a month, and every dose-response impression formed before then is formed on incomplete exposure.

Reasons interact. Cost that would be tolerable alone becomes decisive alongside a tolerability issue that would also have been tolerable alone. Single-reason coding cannot represent this and almost all coding is single-reason.
